Overview

Detalimogene coraplasmid is a non-viral, plasmid-based gene therapy developed by enGene for the localized treatment of bladder cancer, specifically high-risk non-muscle invasive bladder cancer (NMIBC). The drug consists of a DNA plasmid delivered via a non-viral vector system designed for intravesical administration. It encodes two distinct immune-stimulating components: Interleukin-12 (IL-12) to drive adaptive T-cell responses and a RIG-I agonist to activate the innate immune system. By inducing the expression of these proteins directly within the bladder wall, detalimogene coraplasmid aims to generate a robust anti-tumor immune environment while avoiding the systemic toxicities often associated with cytokine therapy. It is currently being evaluated as a potential best-in-class, bladder-sparing alternative for patients who have failed or are unresponsive to Bacillus Calmette-Guérin (BCG) therapy.
