DeTIL-0255 is an autologous adoptive cell therapy consisting of tumor-infiltrating lymphocytes (TILs) derived from a patient’s own tumor, expanded ex vivo with recombinant interleukin-2 and the small molecule CBL-B inhibitor NX-0255. The therapy is designed to enhance the anti-tumor activity of T cells by inhibiting Casitas B-lineage lymphoma proto-oncogene B (CBL-B), an E3 ubiquitin ligase that negatively regulates T cell activation. By blocking CBL-B during the expansion phase, DeTIL-0255 aims to produce a superior TIL product with increased stem-like phenotype and persistence, potentially leading to improved clinical outcomes compared to conventional TIL therapies. It is intended as a single administration following non-myeloablative chemotherapy for patients with advanced gynecological malignancies such as ovarian cancer, cervical cancer, and endometrial cancer who have exhausted standard treatment options[1][2][3][6].
