Deucravacitinib is an oral, small molecule drug used to treat adults with moderate to severe plaque psoriasis. It acts as a highly selective allosteric inhibitor of tyrosine kinase 2 (TYK2), a member of the Janus kinase family. By binding to the regulatory domain of TYK2, deucravacitinib blocks signaling pathways mediated by cytokines such as interleukin-23 (IL-23), interleukin-12 (IL-12), and type I interferons, which are implicated in the pathogenesis of psoriasis. This selectivity distinguishes it from other JAK inhibitors and contributes to its safety profile.
