Drug intelligence / Profile preview
PHVS719 XR (also known as PHVS719) is an investigational, once-daily, oral extended-release tablet formulation of deucrictibant, a highly potent, selective, and competitive small-molecule antagonist of the bradykinin B2 receptor. Developed by Pharvaris, the drug is designed for the long-term prophylactic prevention of hereditary angioedema (HAE) and acquired angioedema (AAE-C1INH) attacks. By blocking bradykinin signaling through the B2 receptor, PHVS719 XR prevents the increased vascular permeability, fluid leakage, and localized swelling that characterize angioedema attacks. The extended-release formulation is optimized to maintain sustained therapeutic plasma concentrations of deucrictibant for over 24 hours, enabling once-daily dosing. PHVS719 XR has demonstrated positive results in the pivotal Phase 3 CHAPTER-3 trial for HAE prophylaxis and is being evaluated in the Phase 3 CREAATE trial for acquired angioedema.
