Deunoxavir marboxil (ADC189) is an oral antiviral drug developed for the treatment of influenza A and B virus infections. It acts as a cap-dependent nucleic acid endonuclease inhibitor, specifically targeting the polymerase acidic (PA) protein of the influenza virus. By inhibiting this enzyme, ADC189 blocks viral mRNA transcription and replication, thereby suppressing both influenza A and B viruses. Preclinical studies have shown that ADC189 has potent antiviral activity against various strains of influenza—including H1N1, H3N2, and highly pathogenic avian influenza—and demonstrates superior efficacy to oseltamivir in animal models. The drug is rapidly metabolized to its active form (ADC189-I07), has a long half-life allowing for single-dose administration, shows no significant food effect on bioavailability, and exhibits a favorable safety profile with minimal central nervous system side effects. Clinical trials have reached Phase III/NDA status in China[1][2][3][4][5][7][8].
