SPL-017 is a deuterated analogue of N,N-dimethyltryptamine (DMT), a naturally occurring psychedelic compound. Developed by Small Pharma (now a subsidiary of Cybin), SPL-017 is part of a research program aimed at optimizing the pharmacokinetic properties of DMT for therapeutic use. DMT is characterized by rapid metabolism by monoamine oxidase A (MAO-A), which results in a very short duration of action when administered intravenously. By replacing hydrogen atoms with deuterium at the alpha-carbon position (d2-DMT), SPL-017 is designed to slow this metabolic degradation, potentially extending the duration of the psychedelic experience and providing a more consistent therapeutic window. The compound acts as a potent agonist at the serotonin 5-HT2A receptor, a mechanism believed to facilitate rapid neuroplasticity and provide antidepressant effects. It is primarily being explored for the treatment of Major Depressive Disorder (MDD) as a fast-acting intervention administered alongside psychological support.
