This is a combination therapy consisting of three distinct drugs: - **Dexamethasone** is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressant properties. It acts primarily by binding to the glucocorticoid receptor, modulating gene expression, and suppressing immune responses. - **Eltrombopag** is an oral small molecule thrombopoietin receptor agonist that stimulates platelet production by activating the thrombopoietin receptor (MPL) on megakaryocyte precursors in the bone marrow. - **Rituximab** is a chimeric monoclonal antibody targeting CD20 on B lymphocytes, leading to B-cell depletion via multiple mechanisms including complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and induction of apoptosis[1][5]. This combination may be used off-label or in clinical trials for conditions such as refractory immune thrombocytopenia (ITP), where synergistic effects are sought from immunosuppression (dexamethasone), stimulation of platelet production (eltrombopag), and targeted B-cell depletion (rituximab). Each component has distinct mechanisms contributing to overall efficacy.
