Overview

This is a combination chemotherapy regimen consisting of dexamethasone (a synthetic glucocorticoid corticosteroid), floxuridine (an antimetabolite and pyrimidine analog), and mitomycin c (an antitumor antibiotic and alkylating agent). The combination has been studied primarily for hepatic arterial infusion (HAI) in patients with unresectable liver metastases from colorectal carcinoma and intrahepatic cholangiocarcinoma. Dexamethasone is included to reduce hepatic toxicity associated with floxuridine. Floxuridine inhibits DNA synthesis by acting as a thymidylate synthase inhibitor, while mitomycin c cross-links DNA strands, inhibiting DNA synthesis and function. This regimen has demonstrated high response rates but also increased biliary toxicity when high-dose mitomycin c is used[1][2][7][9].
