Drug intelligence / Profile preview
Dexamethasone DDS (AGN-201508) is a biodegradable, sustained-release intraocular implant developed by Allergan (now AbbVie) for the treatment of ocular inflammation. The system utilizes the Novadur technology platform, consisting of a solid polymer matrix composed of poly(lactic-co-glycolic acid) (PLGA) that encapsulates dexamethasone, a potent synthetic glucocorticoid. Upon administration, the PLGA matrix slowly degrades into lactic acid and glycolic acid, providing a controlled release of the drug over several weeks to months. While the technology is most widely commercialized under the brand name Ozurdex for posterior segment conditions such as diabetic macular edema, uveitis, and retinal vein occlusion, Allergan also evaluated the Dexamethasone DDS in Phase 1 clinical trials for the management of anterior segment inflammation and pain following cataract extraction. The mechanism of action involves dexamethasone binding to the glucocorticoid receptor, which modulates gene expression to inhibit the synthesis of inflammatory mediators, suppress leukocyte migration, and reduce vascular permeability.
