Overview
Dexamfetamine sulfate is a small molecule central nervous system stimulant and the dextrorotatory isomer of amphetamine. It primarily acts as an indirect sympathomimetic agent by increasing the synaptic concentrations of dopamine and norepinephrine through the inhibition of their respective transporters (DAT and NET) and promoting their release from presynaptic neurons. While traditionally indicated for the treatment of attention-deficit/hyperactivity disorder (ADHD) and narcolepsy, it is being investigated in the Phase 2 DEXAD trial (led by Prof. Dr. Frank Behrens) as an adjunctive therapy to SSRIs or SNRIs for adults with ADHD and comorbid moderate to severe major depressive disorder (MDD). The trial evaluates both immediate-release (IR) and extended-release (XL) formulations to address cognitive symptoms and treatment-resistant depressive features in this specific patient population.
