Overview

This combination therapy consists of the synthetic cannabinoid derivative dexanabinol (ETS2101) administered alongside the standard-of-care chemotherapy doublet, nab-paclitaxel and gemcitabine. Dexanabinol is a non-psychotropic, non-competitive NMDA receptor antagonist that also inhibits TNF-alpha and NF-kappaB signaling. In the context of oncology, dexanabinol is hypothesized to act as a chemosensitizer, potentially enhancing the efficacy of cytotoxic agents by modulating the tumor microenvironment and reducing inflammatory signaling that contributes to resistance. nab-Paclitaxel is an albumin-bound formulation of paclitaxel that promotes microtubule assembly and prevents disassembly, while gemcitabine is a nucleoside analog that inhibits DNA synthesis. This triplet regimen has been primarily investigated in clinical trials for the treatment of advanced or metastatic pancreatic ductal adenocarcinoma (PDAC).
