DexaSite (ISV-305) is an ophthalmic formulation of the potent corticosteroid dexamethasone (0.1%) utilizing the proprietary DuraSite drug delivery platform. Developed by InSite Vision, the DuraSite technology consists of a synthetic mucoadhesive polymer (polycarbophil) that increases the residence time of the active drug on the ocular surface, thereby enhancing bioavailability and potentially allowing for less frequent dosing compared to conventional ophthalmic solutions. Dexamethasone acts as an agonist of the glucocorticoid receptor, leading to the modulation of gene expression that inhibits the production of pro-inflammatory cytokines and reduces capillary permeability and edema. DexaSite was primarily investigated in preclinical studies for the treatment of ocular inflammation and blepharitis.
