Dexo(B16+pIC) is an experimental exosome-based cancer vaccine developed by researchers at the Swiss Federal Institute of Technology Lausanne (EPFL). It consists of exosomes derived from dendritic cells (DCs) that have been pulsed with oxidized B16F10 melanoma cell lysates and matured using the Toll-like receptor 3 (TLR3) ligand poly(I:C). This formulation is designed to enhance the immunogenicity of dendritic cell-derived exosome (Dexo) vaccines, which previously showed limited efficacy in clinical trials due to a lack of pro-inflammatory danger signals. By incorporating both a broad spectrum of tumor antigens and the TLR3 agonist as an adjuvant, Dexo(B16+pIC) promotes the activation of CD4+ and CD8+ T cells, increases the infiltration of NK and NKT cells into the tumor microenvironment, and reduces the frequency of exhausted PD-1+ T cells. Preclinical studies in mouse models of melanoma have demonstrated that intradermal administration of Dexo(B16+pIC) can reduce tumor growth, limit metastasis, and prolong survival.
