Overview

Dexrazoxane is a small molecule cytoprotective agent primarily used to prevent and reduce the incidence and severity of cardiomyopathy associated with doxorubicin administration in women with metastatic breast cancer. It is also indicated for the treatment of tissue damage caused by extravasation of anthracycline chemotherapy agents. Dexrazoxane is a cyclic derivative of EDTA that acts as an intracellular iron chelator, interfering with iron-mediated free radical generation believed to contribute to anthracycline-induced cardiotoxicity. Additionally, it may exert its cardiosparing effects through inhibition of topoisomerase IIβ. Dexrazoxane was discovered in 1972 and has been designated as an orphan drug for these indications.
