Overview

This is a combination of two small molecule drugs, dexrazoxane and mitoxantrone, used in oncology protocols. Dexrazoxane acts as a cytoprotective agent that prevents or reduces anthracycline-induced cardiotoxicity by chelating iron and inhibiting the formation of toxic iron-anthracycline complexes. It also inhibits topoisomerase II, contributing to its cytotoxic effects. Mitoxantrone is a synthetic anthracenedione antitumor antibiotic that intercalates into DNA and inhibits topoisomerase II, leading to DNA strand breaks and inhibition of DNA repair. It has immunosuppressive properties and is structurally related to doxorubicin but was developed to reduce cardiotoxicity compared with traditional anthracyclines[3][4][5]. The combination may be used in pediatric or adult oncology regimens where both agents are indicated—typically when there is concern for cardiac toxicity from cumulative anthracycline exposure[2][5].
