Overview

The combination of dexrazoxane and trastuzumab is an important therapeutic approach in oncology, particularly for patients with HER2-positive breast cancer who are at risk of cardiotoxicity from treatment. Dexrazoxane is a cardioprotective agent that helps prevent heart damage when used alongside anthracycline chemotherapy. Trastuzumab is a monoclonal antibody that targets HER2-positive cancer cells. When used together, this combination aims to provide effective cancer treatment while reducing cardiac side effects. The cardioprotective effect of dexrazoxane is particularly valuable when trastuzumab is administered following or alongside anthracycline-based chemotherapy, as this combination can increase the risk of cardiac toxicity. Studies have shown that dexrazoxane can reduce the risk of clinical heart failure and cardiac events in breast cancer patients undergoing anthracycline chemotherapy with or without trastuzumab. Clinical evidence indicates that patients who received dexrazoxane prior to doxorubicin during chemotherapy maintained higher Left Ventricular Ejection Fraction (LVEF) values throughout trastuzumab treatment compared to those who did not receive dexrazoxane, demonstrating the lasting cardioprotective effect. Importantly, studies have shown that dexrazoxane does not interfere with the anticancer effects of trastuzumab or anthracyclines. While there were historical concerns about dexrazoxane potentially reducing the antitumor activity of anthracyclines or increasing the risk of secondary malignancies, more recent evaluations have found no significant negative impact on cancer outcomes when dexrazoxane is used as a cardioprotective agent.
