Drug intelligence / Profile preview
Dextran-conjugated saroglitazar (also known as Dex-Saro or D-PPAR) is an investigational polymer-drug conjugate consisting of the dual PPARα/γ agonist saroglitazar covalently linked to a dextran nanocarrier via an esterase-cleavable carboxylester bond. Developed by researchers at the University of Illinois Urbana-Champaign, this nanomedicine is designed to selectively target pro-inflammatory macrophages within visceral adipose tissue depots. By delivering the PPARα/γ agonist directly to adipose tissue macrophages, the conjugate modulates macrophage phenotype, reduces systemic inflammation, and improves lipid and glucose metabolism. In preclinical models of diet-induced obesity, treatment with dextran-conjugated saroglitazar led to significant weight loss, improved glucose tolerance, reduced adipose tissue weight, and ameliorated hepatic steatosis without the typical side effects associated with systemic PPAR agonist administration.
