Drug intelligence / Profile preview
Dextran-docosahexaenoic acid-cabazitaxel (dextran-DHA-CTX) is an experimental dual drug conjugate developed to enhance the therapeutic profile of cabazitaxel, a second-generation taxane used for metastatic castration-resistant prostate cancer. The conjugate is synthesized by covalently linking docosahexaenoic acid (DHA), an omega-3 fatty acid, to cabazitaxel and subsequently attaching this complex to a bifunctionalized dextran polymer backbone. This design aims to improve the water solubility of cabazitaxel and facilitate tumor-selective accumulation through the enhanced permeability and retention (EPR) effect. Preclinical evaluations in xenografted nude mice bearing PC-3 and DU145 prostate cancer cells demonstrated that while the conjugate improves solubility and antitumor activity compared to free cabazitaxel, it is less potent than its gamma-linolenic acid (GLA) counterpart, dextran-GLA-CTX. The research was conducted by investigators at Shandong University in collaboration with Santolecan Pharmaceuticals.
