DF1001 is a first-in-class Tri-specific NK cell Engager Therapy (TriNKET) developed by Dragonfly Therapeutics. It is designed to target HER2-expressing tumor cells while simultaneously engaging CD16 and NKG2D receptors on Natural Killer (NK) cells and activated T cells. This dual engagement of NK cells aims to stimulate a potent anti-tumor immune response, including direct lysis of cancer cells and the production of inflammatory cytokines. DF1001 is currently being investigated in Phase I/II clinical trials for the treatment of various HER2-positive advanced solid tumors, including non-small cell lung cancer, metastatic breast cancer, and gastric cancers, both as a monotherapy and in combination with checkpoint inhibitors or chemotherapy.
