Overview

DF5112 is a novel, investigational, long-acting interleukin-12 (IL-12) cytokine therapy developed by Dragonfly Therapeutics. It is engineered using a proprietary cytokine platform designed to extend the protein's half-life and improve its therapeutic index compared to standard recombinant IL-12. While IL-12 is traditionally a potent stimulator of Th1 immune responses and Natural Killer (NK) cell activity—making it a target of interest in oncology—Dragonfly is developing DF5112 to address unmet needs in immune-mediated, autoimmune, and inflammatory diseases. By modulating the IL-12 pathway, the drug aims to restore immune balance or achieve therapeutic effects with a manageable safety profile. DF5112 is currently being evaluated in a Phase 1 first-in-human clinical trial to assess its safety, pharmacokinetics, and pharmacodynamics in healthy volunteers, marking a significant expansion of the company's pipeline beyond oncology.
