Drug intelligence / Profile preview
DFL23806 is a selective small molecule agonist of G-protein coupled receptor 120 (GPR120, also known as free fatty acid receptor 4 or FFAR4) developed by Dompé Farmaceutici [PP0796]. It is designed to activate both G-protein dependent and independent signaling (β-arrestin pathway) of the receptor with high metabolic stability, allowing it to reach the ileum and colon at optimal concentrations [PP0796]. DFL23806 delays receptor internalization, leading to prolonged signal transduction and amplified signaling effects [PP0796]. Preclinical studies in murine models of acute and chronic colitis (induced by DSS and TNBS) have demonstrated that DFL23806 exerts immunomodulatory, anti-inflammatory, and anti-fibrotic effects, improves disease activity index and endoscopic scores, and modulates gut microbiota composition to rescue intestinal dysbiosis [PP0796]. It is being developed as a potential therapeutic approach for inflammatory bowel disease (IBD) and metabolic diseases [PP0796].
