Overview

DFP-10917 + venetoclax is an investigational combination therapy studied for the treatment of relapsed or refractory acute myeloid leukemia (AML). - **DFP-10917** is a novel deoxycytidine nucleoside analog that demonstrates anti-leukemic effects by inducing DNA strand breaks, causing S-phase or G2/M arrest, and leading to apoptotic cell death. Its mechanism is distinct in that short high-concentration exposure leads to S-phase arrest, while prolonged low-dose exposure results in DNA fragmentation and apoptosis. - **Venetoclax** is a selective oral small molecule inhibitor of B-cell lymphoma 2 (BCL-2), an anti-apoptotic protein, thereby promoting programmed cell death in cancer cells. Phase I/II studies are underway to evaluate the safety and preliminary efficacy of the combination, with DFP-10917 administered as a 14-day continuous IV infusion every 28 days and venetoclax as a 14-day oral regimen following a dose ramp-up. The goal is to enhance anti-leukemic activity in patients with relapsed or refractory AML[7].
