DFV890 + MAS825 is an experimental combination therapy comprising two separate investigational immunomodulatory agents developed by Novartis. DFV890 is a potent, selective, orally administered small molecule inhibitor of the NLRP3 inflammasome, which blocks NLRP3-mediated activation of caspase-1 and the subsequent release of inflammatory cytokines including interleukin-1β (IL-1β) and interleukin-18 (IL-18). It is designed to reduce systemic inflammation associated with a range of diseases. MAS825 is a bispecific monoclonal antibody targeting both IL-1β and IL-18, key pro-inflammatory cytokines. MAS825 is administered as a subcutaneous or intravenous infusion and provides dual blockade of these inflammatory mediators. This combination is being studied for additive or synergistic anti-inflammatory effect, especially in patients with coronary heart disease and clonal hematopoiesis of indeterminate potential (CHIP), and has also been investigated in other conditions driven by chronic inflammation.
