Overview

DG041 is a novel, first-in-class, orally administered small molecule developed as a selective and potent antagonist of the prostaglandin E2 EP3 receptor. It was designed for the prevention of arterial thrombosis and its complications, particularly in conditions such as peripheral artery disease. By blocking the EP3 receptor, which is associated with increased risk of vascular diseases, DG041 inhibits PGE2-facilitated platelet aggregation without increasing bleeding risk. The drug demonstrates antiplatelet and antithrombotic effects by reducing platelet activation markers (such as VASP phosphorylation and p-selectin) and inflammatory markers (CRP, MCP1, sICAM). Preclinical studies show that it has good plasma stability but a short half-life in vivo[1][2][3][4][5][6].
