DG071 is a novel small-molecule allosteric modulator of phosphodiesterase 4 (PDE4), specifically targeting the PDE4D isoform. Developed by deCODE genetics, it is intended for the treatment of Alzheimer's disease and other cognitive disorders, including Huntington's disease, schizophrenia, and ADHD. Unlike traditional PDE4 inhibitors that target the catalytic active site and are often limited by emetic side effects such as nausea and vomiting, DG071 binds to a unique allosteric site within the PDE4 regulatory domain. This mechanism allows for the modulation of enzyme activity with a significantly wider safety margin. The compound demonstrated improvements in learning and memory in preclinical models and received FDA clearance to enter Phase I clinical trials in 2008.
