DGAT1 shRNA is a short hairpin RNA (shRNA) designed to silence the expression of diacylglycerol acyltransferase 1 (DGAT1). DGAT1 is an enzyme that catalyzes the final step of triglyceride synthesis by converting diacylglycerol (DAG) to triglycerides. In the context of glioblastoma (GBM), research has shown that radioresistant GBM cells upregulate DGAT1 to maintain lipid homeostasis and evade mitochondrial lipotoxicity. By knocking down DGAT1, this shRNA reduces lipid droplet formation and sensitizes GBM cells to ionizing radiation, both in vitro and in vivo. This suggests that targeting the DGKB-DGAT1 axis is a potential therapeutic strategy for overcoming radioresistance in GBM.
