Overview

DGKζ siRNA is a small interfering RNA (siRNA) designed to silence the expression of diacylglycerol kinase zeta (DGKζ), a type IV lipid kinase. DGKζ plays a critical role in lipid metabolism by phosphorylating diacylglycerol (DAG) into phosphatidic acid (PA), thereby modulating signaling pathways involved in cell growth and survival. In human glioblastoma (GBM), DGKζ is significantly overexpressed compared to precancerous tissues and is regulated by miR-34a. Treatment with DGKζ siRNA has been shown to inhibit GBM cell proliferation, arrest the cell cycle, promote apoptosis, and reduce tumorigenicity in both in vitro and in vivo models. This suggests that DGKζ is a viable therapeutic target for the treatment of glioblastoma and potentially other cancers like colorectal cancer.
