DGKAI

DGKAI is a **small molecule inhibitor** of **diacylglycerol kinase alpha (DGKα)**, an enzyme that converts diacylglycerol (DAG) to phosphatidic acid, modulating signaling pathways that impact cell proliferation, apoptosis, and immune function. In preclinical cancer models, DGKAI suppresses tumor growth by dual mechanisms: (1) inhibiting cancer cell proliferation through targeting DGKα activity and (2) activating anti-tumor immune responses, particularly enhancing effector functions of CD8+ T cells and dendritic cell infiltration. DGKAI shows synergistic anti-tumor activity when combined with immune checkpoint inhibitors targeting PD-L1, suggesting it may augment the efficacy of immunotherapies.
