DGKB shRNA is a short hairpin RNA (shRNA) construct designed to silence the expression of diacylglycerol kinase beta (DGKB) through RNA interference. DGKB is an enzyme that catalyzes the phosphorylation of diacylglycerol (DAG) to produce phosphatidic acid (PA), playing a critical role in lipid signaling and metabolism. In glioblastoma (GBM) research, DGKB shRNA is utilized to model the downregulation of DGKB observed in radioresistant tumor cells. Knockdown of DGKB leads to the accumulation of DAG and a reduction in fatty acid oxidation, which helps GBM cells evade mitochondrial lipotoxicity and maintain lipid homeostasis under stress. This tool is primarily used in preclinical studies to investigate the DGKB-DGAT1 axis as a therapeutic target for overcoming radioresistance in aggressive brain tumors.
