Overview

DH348 is a selective small molecule inhibitor of Carbonic Anhydrase IX (CAIX), an enzyme frequently overexpressed in hypoxic solid tumors that contributes to the acidification of the extracellular microenvironment. By blocking CAIX activity, DH348 helps to neutralize tumor acidity, thereby reducing the stimulus for metastasis and reversing the immunosuppressive effects of a low-pH environment. Preclinical research, notably from the Moffitt Cancer Center and Maastricht University, has demonstrated that DH348 can suppress metastasis in prostate cancer models and sensitize pancreatic cancer models to PD-1 blockade, suggesting its potential as a therapeutic agent to overcome resistance to immunotherapy.
