dHER2 + AS15 is an investigational antigen-specific cancer immunotherapeutic (ASCI) developed by GSK for the treatment of HER2-overexpressing breast cancer. It consists of a recombinant truncated HER2 protein (dHER2) combined with the AS15 Adjuvant System. The dHER2 protein includes the extracellular domain and a portion of the intracellular domain of the HER2 receptor. The AS15 adjuvant is a liposomal formulation containing three immunostimulants: monophosphoryl lipid A (MPL), QS-21 (a saponin), and a CpG oligodeoxynucleotide. The vaccine is designed to induce both humoral (antibody) and cellular (T-cell) immune responses against HER2-positive tumor cells. Clinical development has focused on metastatic breast cancer (often in combination with lapatinib or as maintenance after trastuzumab) and early-stage breast cancer in the adjuvant setting.
