Overview

DHODH-IN-1 (also known as compound 18d) is a potent, small molecule inhibitor of the mitochondrial enzyme dihydroorotate dehydrogenase (DHODH), which plays a critical role in the de novo pyrimidine biosynthesis pathway. It exhibits high inhibitory activity with an IC50 of 25 nM. In vitro studies have demonstrated that DHODH-IN-1 strongly suppresses the proliferation of Jurkat cells (a T-cell leukemia line) and effectively blocks the replication of the measles virus. Despite its high potency in cellular assays, the compound is characterized by a relatively short in vivo half-life of approximately 27 to 41 minutes. DHODH-IN-1 is currently utilized exclusively as a research tool and is not associated with any commercial therapeutic development or clinical trials for human use.
