Overview

DI-B4 is an anti-CD19 monoclonal antibody developed for the treatment of B-cell malignancies. It targets CD19, a protein commonly expressed on the surface of B-cells, including malignant B-cells found in non-Hodgkin lymphoma (NHL), chronic lymphocytic leukemia (CLL), and other B-cell malignancies. The drug works primarily through antibody-dependent cell cytotoxicity (ADCC), recruiting the patient's immune cells to attack and kill cancer cells that express the CD19 protein. DI-B4 contains low levels of fucose, which contributes to its enhanced ADCC activity. It has undergone Phase I clinical trials (NCT01805375) for patients with advanced CD19-positive indolent B-cell malignancies, including relapsed or refractory indolent B-cell lymphoma and chronic lymphocytic leukemia. Results from a Phase I trial showed some patients had partial response or stable disease, with mostly mild side effects.
