DIA-12.3 is a fully human IgG1 monoclonal antibody targeting the N-terminal domain of carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1), an emerging immune checkpoint overexpressed in various solid tumors. Developed by Diatheva in collaboration with the University of Urbino and City of Hope, DIA-12.3 blocks the homophilic and heterophilic interactions of CEACAM1 that suppress immune cell activity. By inhibiting this checkpoint, DIA-12.3 restores and enhances natural killer (NK) cell-mediated cytotoxicity and T-lymphocyte activation against tumor cells. Preclinical studies have demonstrated its efficacy in solid tumor models, including melanoma, bladder cancer, non-small cell lung cancer (NSCLC), and pancreatic ductal adenocarcinoma (PDAC), without inducing neutrophil activation.
