Overview

DIA 3.2-3 is a novel, fully human IgG4 monoclonal antibody targeting CD99, a transmembrane glycoprotein highly expressed on leukemic blasts, leukemic stem cells (LSCs), and Ewing's sarcoma cells. Developed by Diatheva, DIA 3.2-3 is designed to treat hematological malignancies, including acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL), as well as Ewing's sarcoma and myelodysplastic syndromes (MDS). The antibody combines direct tumor cell killing with modulation of the tumor microenvironment while selectively sparing healthy cells. Preclinical studies demonstrate that DIA 3.2-3 undergoes rapid internalization via a non-canonical macropinocytosis-like pathway dependent on lipid rafts, leading to lysosomal sorting, downregulation of membrane proteins, and induction of ribosomal and proteotoxic stress, particularly in FLT3-ITD mutated AML cells.
