Overview

The diabetes-suppressive dendritic cell vaccine is an autologous cell-based immunotherapy designed to treat type 1 diabetes mellitus (T1DM). It consists of a patient's own monocyte-derived dendritic cells that are modified ex vivo with a mixture of phosphorothioate-modified antisense oligonucleotides targeting the primary transcripts of the co-stimulatory molecules CD40, CD80, and CD86. This silencing process renders the dendritic cells tolerogenic (immunosuppressive), meaning they can suppress the autoimmune response against pancreatic beta cells rather than stimulating it. Developed at the University of Pittsburgh and licensed to DiaVacs, Inc., the vaccine is administered intradermally at sites near the pancreas to induce immune tolerance and potentially preserve residual beta-cell function in patients with established or new-onset T1DM.
