Drug intelligence / Profile preview
diABZI3 (also known as diABZI-3 or diABZI STING agonist-1) is a potent, selective, non-nucleotide small-molecule agonist of the Stimulator of Interferon Genes (STING) pathway. Originally developed by GSK, diABZI3 is a dimeric amidobenzimidazole compound designed to mimic the activity of cyclic dinucleotides (CDNs) like cyclic GMP-AMP (cGAMP) but with improved cellular permeability and stability. By binding to STING, diABZI3 induces the phosphorylation of TBK1 and IRF3, leading to the robust production of type I interferons (such as IFN-beta) and proinflammatory cytokines. It has been investigated preclinically for its immunotherapeutic potential in various cancers (such as colorectal cancer), viral infections (including SARS-CoV-2), and parasitic diseases like schistosomiasis, where it helps modulate host immune responses and reduce immunopathology.
