DIACC2010

**DIACC2010** is a first-in-class small molecule developed by Diaccurate as a next-generation payload for antibody-drug conjugates (ADCs) targeting solid tumors and hematologic malignancies. It selectively inhibits **KIF20A** (also known as MKlp2), a kinesin-6 family motor protein essential for cytokinesis, intracellular vesicular trafficking from the Golgi apparatus, and cancer cell proliferation. Preclinical data demonstrate potent cytotoxic activity (median IC50 40 nM) against AML cell lines and primary samples, superior in vivo survival benefits over cytarabine in xenograft models, Golgi scattering confirming target engagement, G2/M cell cycle arrest, and caspase-dependent cell death without toxicity to normal cells like PBMCs or hepatocytes. Its bystander effect and favorable profile support improved ADC therapeutic index.[1][3]
