Overview

Diacerein is a small molecule anthraquinone derivative with anti-inflammatory and anti-rheumatic properties. It acts primarily as an inhibitor of interleukin-1 beta (IL-1β) production and activity by reducing the synthesis of IL-1 converting enzyme, thereby suppressing downstream signaling pathways involved in inflammation. Diacerein is classified as a symptomatic slow-acting drug for osteoarthritis (SYSADOA), used mainly to treat degenerative joint diseases such as osteoarthritis. Its effects typically become apparent after 2–4 weeks of administration and are more pronounced after 4–6 weeks. The drug also demonstrates antinociceptive effects through inhibition of glutamatergic neurotransmission and cytokine signaling[1][3][4][5].
