Overview

Dianicline is a small molecule drug developed by Sanofi-Aventis as a selective partial agonist of neural nicotinic acetylcholine receptors, primarily targeting the α4β2 subtype. Its mechanism of action involves modulating these receptors to reduce nicotine dependence and assist in smoking cessation. Dianicline was designed to mimic some effects of nicotine while reducing withdrawal symptoms and cravings, with the aim of supporting individuals attempting to quit smoking. It is structurally and pharmacologically similar to varenicline but may have an improved side effect profile. Despite initial promise in early-phase clinical trials, development was discontinued after unfavorable results in Phase III studies[1][5][7].
