Overview

DiaPep277 is a synthetic 24-amino acid peptide derived from the human heat shock protein 60 (Hsp60, residues 437–460). It was developed as an immunomodulatory therapy for type 1 diabetes mellitus and other autoimmune diseases. The drug acts by modulating the immune response, specifically promoting anti-inflammatory effects and shifting cytokine secretion away from a pro-inflammatory profile through interaction with Toll-like receptor 2 (TLR2). This mechanism aims to preserve pancreatic beta-cell function by reducing autoimmune destruction. Clinical trials demonstrated that DiaPep277 is safe and may help maintain endogenous insulin production in some patients with recent-onset type 1 diabetes, though efficacy results were modest. Development was halted after issues related to phase III trial data integrity[1][3][5][6][8].
