Overview

Diazoxide is a small molecule, nondiuretic benzothiadiazine derivative used primarily to manage hypoglycemia due to hyperinsulinism. It acts by opening ATP-sensitive potassium channels on pancreatic beta cells, leading to membrane hyperpolarization and inhibition of insulin release. This results in increased blood glucose levels. Diazoxide is also used intravenously for hypertensive emergencies as a vasodilator and has been studied for use in Prader-Willi syndrome (as diazoxide choline) to treat hyperphagia. It is chemically related to thiazide diuretics but lacks significant diuretic activity[1][3][5].
