Overview

This is a combination drug consisting of diclofenac sodium, a nonsteroidal anti-inflammatory drug (NSAID), and thalidomide, an immunomodulatory agent with anti-inflammatory and anti-angiogenic properties. Diclofenac sodium acts primarily by inhibiting cyclooxygenase (COX) enzymes, reducing the synthesis of prostaglandins involved in pain and inflammation. Thalidomide modulates the immune response by inhibiting tumor necrosis factor-alpha (TNF-α) production and affecting other cytokines; it also has anti-angiogenic effects. While both drugs are individually approved for various indications—diclofenac for pain/inflammation in conditions like arthritis, and thalidomide for multiple myeloma and erythema nodosum leprosum—their combination is not widely marketed or approved as a fixed-dose product. Limited preclinical research has explored their combined use in settings such as skin allograft survival, but there is no evidence of broad clinical adoption or regulatory approval for this specific combination[1][9]. Both agents carry significant safety considerations; notably, thalidomide is highly teratogenic.
