Dicyclomine (also known as dicycloverine) is an anticholinergic and antispasmodic small molecule drug primarily used to treat symptoms of irritable bowel syndrome (IBS), particularly intestinal spasms and hypermotility. It acts as a muscarinic receptor antagonist—specifically at the M1, M2, and M3 subtypes—blocking acetylcholine at these sites in the gastrointestinal tract. This leads to relaxation of smooth muscle and reduced cramping. Dicyclomine also non-competitively inhibits bradykinin- and histamine-induced spasms in the gut. The drug was first approved for medical use in 1950 and is available both as a generic medication and under several brand names[1][2][4][5][6][7].
