Overview

Dicycloplatin is a second-generation platinum-based antineoplastic agent developed as a supramolecular complex of carboplatin and 1,1-cyclobutanedicarboxylic acid. It was designed to improve the solubility and stability of platinum drugs while minimizing the severe toxicities, such as nephrotoxicity and ototoxicity, typically associated with cisplatin. The drug exerts its therapeutic effect by binding to DNA, primarily forming intrastrand cross-links at the N7 positions of guanine and adenine residues. These DNA adducts interfere with DNA replication and transcription processes, ultimately triggering cell cycle arrest and apoptosis in rapidly proliferating malignant cells. Dicycloplatin was approved by the National Medical Products Administration (NMPA) of China in 2012 for the treatment of non-small cell lung cancer (NSCLC) and has been investigated for various other solid tumors.
