Overview

Didaxini (also known as EVT-201) is a small molecule partial positive allosteric modulator (PAM) of the GABA-A receptor, specifically targeting the benzodiazepine binding site. Developed by Zhejiang Jingxin Pharmaceutical under license from Evotec, it is indicated for the treatment of insomnia. Unlike traditional full-agonist benzodiazepines or non-benzodiazepine "Z-drugs" (such as zolpidem), didaxini's partial agonism is designed to provide sufficient sedative-hypnotic efficacy while reducing the incidence of common side effects such as residual next-day sedation, cognitive impairment, motor incoordination, and the potential for tolerance or dependence. The drug received approval from China's National Medical Products Administration (NMPA) in 2023, making it a novel therapeutic option for patients with sleep onset and maintenance difficulties.
