Drug intelligence / Profile preview
Didodecyl-methotrexate (ddMTX) is a lipophilic derivative of methotrexate, chemically produced by esterification of methotrexate with dodecyl groups. Designed to enhance drug delivery and cellular uptake, ddMTX is typically formulated with lipid nanoemulsions (LDE-ddMTX) for improved pharmacokinetics and cytotoxicity. Preclinical studies demonstrate that ddMTX has markedly higher cytotoxicity compared to methotrexate when used in nanoparticle formulations, with potential therapeutic applications in cancer and diseases where methotrexate is already used. Increased lipophilicity is intended to optimize passive transport and intracellular concentration, though the clinical use remains investigational.[2]
