Diethylnitrosamine (DEN), also known as N-nitrosodiethylamine, is a potent carcinogenic compound found in various environmental sources such as tobacco smoke, processed meats, cheese, soybean products, water supplies, cosmetics, and some pharmaceutical agents[1][4]. It is not used therapeutically but is widely employed in research to induce hepatocellular carcinoma (HCC) and other tumors in experimental animal models due to its strong hepatocarcinogenic and mutagenic properties[1][3][5]. DEN exerts its effects primarily through metabolic activation that generates high levels of reactive oxygen species (ROS), leading to DNA damage via alkylation and oxidative stress. This results in mutations that drive carcinogenesis[1][5][6]. The compound is especially notable for inducing liver cancer that closely mimics human HCC pathology; thus it serves as a standard agent for preclinical studies of liver cancer mechanisms and therapies[1].
