DIFF-flu is a live-attenuated influenza vaccine (LAIV) candidate developed by Vivaldi Biosciences using its proprietary Delta-NS1 (DELNS1) technology. The vaccine consists of influenza virus strains in which the gene for Non-Structural protein 1 (NS1) has been deleted. NS1 is the primary viral protein responsible for antagonizing the host's innate immune response, specifically by blocking the production of interferon. By removing this gene, the vaccine virus becomes replication-deficient in normal human cells but remains highly immunogenic. Upon intranasal administration, DIFF-flu induces a rapid and robust localized interferon response, which acts as a natural adjuvant to stimulate broad-spectrum immunity, including mucosal IgA, systemic IgG, and T-cell responses. This mechanism is designed to provide superior protection against both homologous and heterologous (drifted) influenza strains compared to traditional inactivated or live-attenuated vaccines.
