Overview

This is a multi-component antimalarial regimen developed by the University of Oxford, combining the artemisinin-based combination therapy (ACT) dihydroartemisinin-piperaquine with the endectocide ivermectin and the 8-aminoquinoline primaquine. The regimen is designed for malaria elimination efforts by simultaneously treating the infection in the human host and preventing further transmission. Dihydroartemisinin and piperaquine provide rapid and sustained clearance of blood-stage parasites. Primaquine is included for its ability to clear Plasmodium falciparum gametocytes and Plasmodium vivax hypnozoites. Ivermectin acts as a transmission-blocking agent by killing Anopheles mosquitoes that feed on treated individuals. A Phase 1 study in healthy Thai adults evaluated the safety, tolerability, and pharmacokinetic interactions of this combination, as well as its mosquito-lethal efficacy.
